No you cannot apply via Singapore, according to this website. I think you have to apply from Myanmar.4. Original and copy of the residence permit (e.g. PR and RE-ENTRY PERMIT, EP) that will be valid for at least 6 months beyond the date of application.

Your visa to Hong Kong is based on your country of citizenship, i.e your Myanmar passport. Whether Hong Kong accepts visa application made outside of country of citizenship/residence i.e Singapore, you have to check with the embassy.
The question is hard for me to understand, as it's not clear what the 'Short term pass' referred to is. Is that it's formal name? What is it's duration, and is it marked as multiple-entry?
